Scientific Researches on Tadalafil
“Urodynamic effects of the combination of tamsulosin and daily tadalafil in the men having lower urinary tract symptoms secondary to benign prostatic hyperplasia: a randomized, placebo-controlled clinical trial” by Rommel Prata Regadas et al evaluated the effects of tamsulosin combined with tadalafil in treatment of lower urinary tract symptoms produced due to presence of benign prostatic hyperplasia in males. This was an urodynamic study in which the combination therapy of tamsulosin with tadalafil was compared with a combination of tamsulosin and placebo in patients who were showing symptoms of lower urinary tract problems that were secondary to benign prostatic hyperplasia. All of the patients were put through to a baseline evaluation of their health and condition of their disease before receiving either of the therapies. The patients were divided into two groups on the basis of the combination therapy the received. The first group comprised of 20 people who were given tamsulosin in a dose of 0.4 mg on a daily basis in a combination of tadalafil in a dose of 5 mg. The second group comprising of 20 patients as well received 0.4mg of tamsulosin along with placebo every day. The remedy was sustained for 30 days after which the patients were exposed to urodynamic study specified for the end. The patients were evaluated for any variations in the urodynamic variables during the phase of voiding and the changes seen in the peak urinary flow rates. The pressure of the detrusor during peak flow was also checked for any changes at the end of the treatment. It was noticed that the pressure of detrusor recorded during peak flow rates had significantly reduced in the group receiving tamsulosin along with tadalafil as noted with the group receiving placebo in place of tadalafil. An increase in maximum flow rates was also observed in the patients of both groups but it was indistinguishable between these groups. An improvement in the voiding score of patients receiving tamsulosin and tadalafil was also seen. The researchers came to a conclusion that the use of tamsulosin along with tadalafil reduced the detrusor pressure without affecting the peak flow rates in patients, therefore, can be used to alleviate lower urinary tract symptoms. 
“Clinical Efficacy of Phosphodiesterase-5 Inhibitor Tadalafil in Eisenmenger Syndrome—A Randomized, Placebo-controlled, Double-blind Crossover Study” by SaibalMukhopadhyay et al discussed the role of tadalafil in treating Eisenmenger Syndrome. It was a double-blind study consisting of a placebo group to compare the effects of tadalafil with this placebo group in treating Eisenmenger syndrome. The parameter set to evaluate the efficacy of either of the agents was the 6 min test in which the patient had to walk. The patients were also to be evaluated for pulmonary vascular resistance, blood flow towards lung, systemic vascular resistance and how the agent affected the systemic oxygen saturation. 28 patients were selected for this purpose who were suffering from Eisenmenger syndrome with weights greater than 30 kg. Some of the patients were given tadalafil in a dose of 40 mg while others were given placebo in the same amount for a duration of 6 weeks. This was followed by a 2 week washout period after which the patients were switched to another drug. All of the patients were evaluated at baseline as well as after 6 weeks and finally at the end of the therapy according to the parameters described above. The study was completed by all of the patients of which those who received tadalafil presented more improvement in the pulmonary vascular resistance as compared to the placebo-treated patients. It was finished that the management of tadalafil in patients tolerating Eisenmenger Syndrome was well suffered and produced visible improvements in the disease as measured by different parameters. 
Description and Mechanism of Action of Tadalafil
Tadalafil does not cause an erection directly but helps the body to maintain an erection indirectly by inhibiting the enzyme phosphodiesterase 5 and maintaining high levels of the chemical, nitric oxide. NO is normally released during stimulation which causes relaxation of the smooth muscles present in the penis. As the smooth muscles relax, more blood flows in the penile organ for a prolonged duration causing a maintained erection.

Use of Tazzle is linked with a number of side effects some of which are common while the others are rare among the patients. These include back aches, pain in muscles and blockage of the nose. It also results in loss of memory in patients sometimes along with flushing of the skin and facial redness. Sneezing and sore throat has also been noticed in some instances. Tazzle can also upset the stomach of the patient using it and can also result in the development of diarrhea. The patient can also suffer from hard ad painful erections sometimes which are unwanted and do not go away easily but this happens rarely. 
Tazzle has a potential to interact with different drugs antibiotics e.g. Clarithromycin. Some of the drugs which must not be taken with drugs like Clotrimazole and Conivaptan. Diclofenac and Doxazosin are also found to cross-react with Tazzle due to the presence of tadalafil in it. Erythromycin, Imatinib and Isoniazid should also not be consumed when a person is already taking Tazzle for ED.Nefazodone and Rifampin are also discouraged to be used by patients taking Tazzle already.
